目的:研究广东舟山眼镜蛇蛇毒的镇痛活性成分,为广东舟山眼镜蛇蛇毒的开发利用,寻找新型镇痛药奠定基础。方法:采用对氨基苯甲酸-Sepharose 6B亲和色谱与Sephadex G-50凝胶色谱进行分离纯化;利用SDS-聚丙烯酰胺凝胶电泳法与高效液相色谱法进行纯度的鉴定;应用激光飞行质谱测定其相对分子质量;采用神经病理性疼痛模型评估产物的镇痛活性。结果:经过两步柱色谱法分离得到的产物为单一成分,高效液相色谱测定相对纯度达到95%;相对分子质量为6741.236 Da;在神经病理性疼痛模型,单次腹腔给药后镇痛作用显著,持续时间超过24 h。结论:广东舟山眼镜蛇毒经过两步柱色谱后得到一高纯度的镇痛活性产物,值得进一步开发研究。
OBJECTIVE: To study the analgesic active ingredients of Cobra snake venom from Zhoushan, Guangdong and lay a foundation for the development and utilization of Cobra snake venom in Zhoushan, Guangdong Province, looking for new analgesics. Methods: Separation and purification of p-aminobenzoic acid-Sepharose 6B affinity chromatography and Sephadex G-50 gel chromatography were performed. Purity was identified by SDS-polyacrylamide gel electrophoresis and high performance liquid chromatography. The relative molecular mass was determined. The neuropathic pain model was used to assess the analgesic activity of the product. Results: The product obtained by two-step column chromatography was a single component. The relative purity was 95% and the relative molecular mass was 6741.236 Da. In the model of neuropathic pain, the analgesic effect after single intraperitoneal administration was significant , Lasting more than 24 h. Conclusion: The cobra venom from Zhoushan, Guangdong, obtained a highly pure analgesic active product after two-step column chromatography, which deserved further research.
