目的 :探讨健胃愈疡颗粒剂治疗胃溃疡的疗效机理。方法 :用Okabe改良法复制大白鼠胃溃疡模型。运用免疫组织化学法和原位杂交检测胃黏膜细胞Bc1 2、Bax基因蛋白和mRNA表达的调节作用。结果 :健胃愈疡颗粒组Bc1 2基因蛋白和mRNA表达强度 7d后明显高于模型组和胃苏颗粒组 ,14d后则无明显差异。Bax基因蛋白和mRNA表达强度 7d后明显高于模型组和胃苏颗粒组 ,与正常组无显著性差异 ,14d后的比值均有所恢复。结论 :健胃愈疡颗粒剂能显著提高胃黏膜细胞的Bc1 2基因蛋白和mRNA的表达强度 ,调节Bc1 2 /Bax的比值 ,使其恢复正常的动态平衡。
Objective: To explore the therapeutic mechanism of Jianweiyuyang granule on gastric ulcer. METHODS: The rat model of gastric ulcer was replicated using the Okabe modified method. Immunohistochemistry and in situ hybridization were used to detect the regulation of protein and mRNA expression of Bcl2 and Bax genes in gastric mucosal cells. Results : The protein and mRNA expression of Bc1 2 gene in Jianweiyuyang granule group was significantly higher than that of model group and Weisu granule group after 7 days, but no significant difference was found after 14 days. Bax gene protein and mRNA expression intensity was significantly higher than that of model group and Weisu granule group after 7 days, and there was no significant difference between the normal group and the normal group. The ratio of Bax gene protein and mRNA recovered after 7 days. Conclusion: Jianweiyuyang granules can significantly increase the expression of Bc1 2 gene protein and mRNA in gastric mucosal cells, regulate the ratio of Bc1 2 /Bax, and restore normal homeostasis.
