杂质依据极性的不同可分为强极性、中等极性和弱极性3类,在不同溶剂中的溶解性能有显著差异。本研究应用三级逆流离心萃取技术纯化可利霉素,考察洗涤条件对不同杂质的去除情况。结果显示,洗涤步骤对3类杂质均有去除效果,但对较强极性杂质的去除更有效。优化洗涤条件,以1%磷酸二氢钠溶液作为洗涤液,在20℃、水酯相比1∶1.5、离心萃取器相对离心力为74×g时,总杂质分离效果较好。除杂率大于50%,较传统的错流萃取工艺提高15%。
Impurities can be divided into three categories according to their polarities: strong polarity, moderate polarity and weak polarity. Their solubility in different solvents is significantly different. In this study, three-stage countercurrent centrifugal extraction technology was used to purify the clotrimixin, and to investigate the removal of different impurities by washing conditions. The results show that the washing step removes all three types of impurities but is more effective at removing more strongly polar impurities. The optimized washing conditions were as follows: 1% sodium dihydrogen phosphate solution was used as washing solution, and the relative centrifugal force of centrifugal extractor was 74 × g at 20 ℃ and water ester ratio was 1: 1.5. Greater than 50% removal of impurities, compared with the traditional cross-flow extraction process increased by 15%.
