目的 考察甘草和五味子对大鼠肝微粒体CYP45 0的诱导作用及其对丙咪嗪体外代谢的影响。方法 大鼠分别给予甘草或五味子的水提物连续 3d或 6d ,然后制备肝微粒体并测定其CYP45 0的水平 ;以丙咪嗪为底物 ,采用体外温孵法 ,用HPLC测定丙咪嗪及其主要代谢物的量以考察其代谢情况。结果 给予中草药的大鼠肝微粒体CYP45 0水平均显著提高 ,其对丙咪嗪的体外代谢速率显著加快。结论 研究显示甘草和五味子对大鼠CYP45 0有诱导作用 ,且与剂量或诱导时间有关
Objective To investigate the effect of licorice and Schisandra chinensis on rat liver microsomal CYP45 0 and its effect on the metabolism of imipramine in vitro. Methods Rats were given water extracts of Glycyrrhiza or Schisandra respectively for 3d or 6d, then liver microsomes were prepared and their CYP450 levels were measured. With imipramine as substrate, in vitro incubation was used to determine imipramine by HPLC. The amount of its major metabolites was examined for its metabolism. Results The level of CYP45 0 in liver microsomes of rats given Chinese herbal medicine was significantly increased, and the in vitro metabolic rate of imipramine was significantly accelerated. Conclusion Studies have shown that licorice and schisandrin have an inducing effect on rat CYP45 0 and are related to dose or induction time.
