目的 研究瘦素和过氧化酶体增殖物激活受体γ(PPAR γ)在大鼠脂肪生长与分解过程中的作用.方法 将雄性离乳Wistar大鼠随机分为对照组(C1组)和高脂喂养组,分别用普通饲料和高脂饲料喂养17周后,高脂组改为普通饲料喂养,同时分为正常量喂养组(C2组),80%量限食喂养组(HR组),80%量限食喂养同时注射瘦素组(HRL组)和正常量喂养同时注射瘦素组(HL组).分别在10周、17周、18周和23周末期测量各组大鼠的体重,取大鼠血液检测瘦素水平,应用免疫组化染色和Western blot检测大鼠腹股沟脂肪组织PPARγ蛋白表达.结果 高脂喂养阶段,大鼠的体重,血液瘦素水平和脂肪组织中PPARγ蛋白的表达都较C1组显著增高.限食喂养阶段,限食使大鼠血液瘦素值显著降低(P<0.05);限食或注射瘦素都使大鼠的体重、脂肪组织中PPARγ蛋白的表达量显著减少(P,<0.05);限食同时注射瘦素时减少更加显著(P<0.05).结论限食同时注射瘦素共同发挥增强脂肪分解代谢的作用,与抑制PPAR γ蛋白的表达相关.“,”Objective To study the role of leptin and peroxisome proliferator-activated receptor ganma (PPAR γ) on the growth and decomposition of fat in rats.Methods Male weaning Wistar rats were randomly divided into control group(C1 group,normal diet) and high fat diet group(high fat diet) for 17 weeks.Then the high fat groups were divided into:normal diet group(C2 group),80% volume limit feeding group(HR group),80% volume limit feeding accompanied by leptin injection group(HRL group) and normal feeding accompanied by leptin injection group(HL group).At the end of 10th,17th,18th and 23rd week,rat weight was measured,leptin level was detected,immunohistochemistry and Western blot were used to detect the expression of the PPAR gamma protein in inguinal adipose tissue.Results In high fat feeding stage,the body weight,blood leptin level and the expression level of PPAR gamma protein in adipose tissue were significantly higher than those in C1 group.The blood leptin levels decreased significantly,the expression level of PPAR gamma protein in adipose tissue and weight of rats decreased significantly by food restriction or leptin injection,but decreased more significantly when food restriction accompanied by leptin injection(P<0.05).Conclusion The dietary restriction and leptin reduced body fat and body weight of rats,which might be related to inhibition of the expression of PPAR gamma protein.
