目的探讨慢性复合应激对大鼠海马神经元中生长休止蛋白7(Gas7)的表达变化及意义。方法36只大鼠随机分为慢性复合应激组和正常对照组。复合应激组动物进行6周的垂直旋转、睡眠剥夺、捆绑(6h/d)和夜间光照等慢性复合性应激试验;实验结束后,所有动物采用免疫组织化学和免疫印迹等方法检测大鼠海马Gas7蛋白表达的变化和神经元凋亡情况。结果Gas7在对照组和实验组大鼠海马各区均有表达,主要表达在海马神经元的胞质和突起内。其中在CA1区和齿状回呈较强阳性表达,CA3区表达则较弱;慢性复合应激组CA1区和齿状回阳性染色加深,平均吸光度明显上升(P<0.05),而CA3区则不明显(P>0.05)。慢性复合应激组中部分切片在下托可见少量Caspase-3免疫反应阳性细胞。免疫印迹检测表明,慢性复合应激组大鼠海马Gas7表达明显增强,与正常对照组相比,差异具有显著性(P<0.05)。结论Gas7可能参与了在应激情况对神经元的保护作用,以及促进神经元的发生和发育。
Objective To investigate the changes and significance of chronic multiple stress on expression of Gas7 in hippocampal neurons of rats. Methods Thirty-six rats were randomly divided into chronic stress group and normal control group. The rats in the composite stress group were subjected to chronic composite stress test such as vertical rotation for 6 weeks, sleep deprivation, bundling (6h / d) and night light. After the experiment, all animals were examined by immunohistochemistry and immunoblotting Change of hippocampal Gas7 protein expression and neuronal apoptosis. Results Gas7 was expressed in the hippocampus in both control and experimental groups, and was mainly expressed in the cytoplasm and protrusions of hippocampal neurons. The CA1 and dentate gyrus showed a strong positive expression in CA1 and a weak expression in CA3, while the CA1 and dentate gyrus in chronic stress group had a higher positive staining and a higher mean absorbance (P <0.05), while CA3 Not obvious (P> 0.05). In the chronic complex stress group, a small amount of Caspase-3 immunoreactive cells were seen in the sections of the basal lamina. Immunoblotting showed that the expression of Gas7 in the hippocampus of chronic multiple stress group was significantly increased compared with the normal control group (P <0.05). Conclusion Gas7 may be involved in the protection of neurons in stress conditions and promote the occurrence and development of neurons.
