目的 :证明血灵对血管平滑肌钙通道阻滞作用。方法 :用累积浓度法依次加入去甲肾上腺素(NE) ,得NE引起的血管平滑肌收缩的量效曲线 ,收缩张力恢复正常后加入血灵 ,测定给药后量效曲线 ,观察血灵对NE引起的血管平滑肌收缩的影响 ;同法观察血灵对KCl、CaCl2 引起的血管平滑肌收缩的影响 ,及对NE引起的依赖于细胞内钙与细胞外钙收缩反应的影响。结果 :血灵能够使NE和CaCl2收缩血管平滑肌的量效曲线下移 ,作用性质与维拉帕米(Ver)相似 ,呈非竞争性拮抗反应 ;血灵对NE引起的依赖于细胞内钙收缩有明显的抑制作用 ,而对外钙收缩则无明显的抑制。结论 :血灵的作用类似于钙通道阻滞剂维拉帕米 ,提示血灵降血压与钙通道阻滞作用有关
Objective: To prove the effect of Xueling on the calcium channel block of vascular smooth muscle. Methods: Norepinephrine (NE) was sequentially added by cumulative concentration method to obtain the dose-response curve of NE-induced vascular smooth muscle contraction. After the contraction tension returned to normal, blood spirit was added to determine the dose-effect curve after administration. The effect of vasoconstriction caused by vascular smooth muscle; the effect of hemoglobin on the contraction of vascular smooth muscle caused by KCl and CaCl2, and the effect of NE on the intracellular calcium and extracellular calcium contraction response. RESULTS: Xueling could decrease the dose-response curve of NE and CaCl2 contraction of vascular smooth muscle, and the nature of the effect was similar to that of Verapamil (Ver), showing a non-competitive antagonistic response; Xueling was dependent on intracellular calcium contraction due to NE. There is a clear inhibitory effect, while no significant inhibition of external calcium contraction. Conclusion : The effect of Xueling is similar to that of the calcium channel blocker verapamil , suggesting that the lower blood pressure is related to the effect of calcium channel blockade
