目的总结细胞自噬在周围神经损伤后病理生理变化过程中的作用及调控机制,为周围神经损伤的治疗提供新的思路。方法广泛查阅近5年与细胞自噬在周围神经损伤及再生方面的相关文献,并进行总结与讨论。结果通过对小鼠进行药物干预及基因敲除等方法调节其自噬功能后证实,雪旺细胞自噬主要通过JNK/c-Jun通路影响后续的髓鞘崩解、炎性细胞浸润及轴突再生等一系列过程,通过调节雪旺细胞自噬可以改变周围神经损伤后瓦勒变性的发生、发展,维持周围神经结构的完整性,但其对后续轴突再生的作用仍存在争议。结论细胞自噬过程在周围神经损伤后病理生理变化过程中起着重要的调节作用,深化研究其机制及作用,可为周围神经损伤后治疗方法的研究提供新思路。
Objective To summarize the role and regulation of autophagy in the pathophysiological changes after peripheral nerve injury, and to provide a new idea for the treatment of peripheral nerve injury. Methods Extensive review of the recent 5 years and cell autophagy in peripheral nerve injury and regeneration related literature, and to summarize and discuss. Results After autophagic function was regulated by drug intervention and gene knockout in mice, it was confirmed that Schwann cell autophagy mainly affected subsequent disintegration of myelin sheath, inflammatory cell infiltration and axons through JNK / c-Jun pathway Regeneration and a series of processes, by regulating Schwann cell autophagy can change the occurrence of Wale degeneration after peripheral nerve injury, development and maintain the integrity of the peripheral nerve structure, but its role in subsequent axonal regeneration is still controversial. Conclusion The autophagy plays an important regulatory role in the pathophysiological changes after peripheral nerve injury. Deeper studies on its mechanism and action may provide new ideas for the treatment of peripheral nerve injury.
