目的探讨正丁酸钠对人肝癌细胞恶性表型的逆转作用。方法以0.2mmol/L的正丁酸钠(sodiumbutyrate,SB)作用于培养的人肝癌细胞GHC-3,动态观察癌细胞生长、形态、ConA凝集性、琼脂集落形成率、染色体数目、DNA合成、DNA含量等表型的变化。结果SB作用7天,人肝癌细胞生长抑制;SB作用15天ConA凝集现象减弱;作用30天后细胞形态由多角型转变为棱形,细胞ConA凝集现象基本消失;经SB作用15天、30天、45天的细胞球脂集落形成率由对照组的4.54%下降为2.53%、1.45%、1.31%;SB作用30天、45天细胞3小时3H-TdR掺入率由2330.24士10.31cpm/105cells下降为1372.5123.41cpm/105cells和1136.23土17.23cpm/105cells,细胞DNA含量由198.511.18μg/107cells下降为134.78土0.53μg/107cells和104.78土0.53μg/107cells;均有显著差异(P<0.01)。染色体主流范围变窄,二倍体比例逐渐升高而亚三倍体比例逐渐下降,三倍体消失。结论人肝癌细胞在正了酸钠持续作用下,某些恶性表型可发生进行性逆转。
Objective To investigate the reversal effect of sodium butyrate on the malignant phenotype of human hepatoma cells. Methods 0.2 mmol/L sodium butyrate (SB) was used to treat human hepatocellular carcinoma cell line GGC-3. The growth, morphology, ConA agglutination, agar colony formation rate, chromosome number, and DNA synthesis were observed dynamically. Changes in phenotypes such as DNA content. Results After SB treatment for 7 days, the growth of human hepatocellular carcinoma cells was inhibited; ConA agglutination was weakened after SB treatment for 15 days; After 30 days, the morphology of cells changed from polygonal to prismatic, and the ConA agglutination of cells basically disappeared; SB effect was 15 days and 30 days. The 45-day cell glomerular colony formation rate decreased from 4.54% in the control group to 2.53%, 1.45%, 1.31%; SB action for 30 days and 45-day cells for 3 hours 3H-TdR incorporation rate by 2330 .24 ± 10.31 cpm/105 cells decreased to 1372.5123.41 cpm/105 cells and 1136.23 soil 17.23 cpm/105 cells. The mainstream of chromosomes narrowed, the proportion of diploids gradually increased and the proportion of sub-triploids gradually decreased, and triploids disappeared. Conclusion Under the sustained action of sodium sulfate, some malignant phenotypes of human hepatocellular carcinoma cells can be progressively reversed.
