目的研究乳腺癌组织中第157位苏氨酸磷酸化p27~(kip1)(P-p27Thr157)的表达情况,探讨其表达与乳腺癌发生发展的关系。方法回顾性收集2004年1月—2005年12月于嘉兴市第一医院行手术切除的111例乳腺癌组织标本以及距肿瘤边缘5 cm以上的癌旁组织,并收集8对新鲜乳腺癌和相应癌旁正常组织标本用于蛋白提取。分别采用免疫组织化学EnVision二步法和蛋白质印迹法检测乳腺癌及相应癌旁正常组织中P-p27Thr157的表达情况;分析其与乳腺癌临床病理特征的关系。结果免疫组织化学和蛋白质印迹结果均显示,P-p27Thr157在乳腺癌组织中的阳性表达显著高于其在癌旁正常组织中的表达,本组乳腺癌组织中P-p27Thr157阳性表达率为87.4%(97/111),高表达者64例(57.7%),低表达者47例(42.3%),而癌旁组织中阳性表达率为9.9%(11/111),2组之间差异有统计学意义(P<0.05);P-p27Thr157表达与肿瘤大小、淋巴结转移情况和TNM分期有关(均P<0.01);P-p27Thr157高表达患者的总生存率低于低表达患者,差异有统计学意义(P<0.05);P-p27Thr157与增殖指标Ki-67表达呈正相关(r=0.528,P<0.05),与p27表达呈负相关(r=-0.458,P<0.05)。结论乳腺癌组织中p27Thr157异常磷酸化可能参与了乳腺癌的发生、发展过程。检测Thr157磷酸化p27对乳腺癌的临床诊断及预后判断具有一定的参考价值。
Objective To investigate the expression of phosphorylated p27 kip1 (P-p27Thr157) in breast cancer tissue and to investigate its relationship with the occurrence and development of breast cancer. Methods Retrospectively collected 111 cases of breast cancer tissue resected from the First Hospital of Jiaxing from January 2004 to December 2005 and the adjacent tissues more than 5 cm from the edge of the tumor and collected 8 pairs of fresh breast cancer and their corresponding Next to normal tissue samples for protein extraction. The expression of P-p27Thr157 in breast cancer tissues and adjacent normal tissues was detected by immunohistochemistry EnVision two-step method and Western blot respectively. The relationship between the expression of P-p27Thr157 and the clinicopathological features of breast cancer was analyzed. Results The results of immunohistochemistry and Western blotting showed that the positive expression of P-p27Thr157 in breast cancer tissues was significantly higher than that in the adjacent normal tissues. The positive rate of P-p27Thr157 expression in breast cancer tissues was 87.4% (97/111), 64 (57.7%) were highly expressed, 47 (42.3%) were low expressed, while the positive rate was 9.9% (11/111) in paracancerous tissues. There was statistical difference between the two groups (P <0.05). The expression of P-p27Thr157 was correlated with tumor size, lymph node metastasis and TNM stage (all P <0.01). The overall survival rate of patients with high expression of P-p27Thr157 was lower than that of patients with low expression (P <0.05). There was a positive correlation between the expression of P-p27Thr157 and proliferation index Ki-67 (r = 0.528, P <0.05) and negative correlation with p27 expression (r = -0.458, P <0.05). Conclusion The abnormal phosphorylation of p27Thr157 in breast cancer may be involved in the occurrence and development of breast cancer. Detection of p27 Thr157 phosphorylation of breast cancer clinical diagnosis and prognosis of a certain reference value.
