目的 :观察补肾益智方对Alzheimer病 (AD)模型大鼠突触的影响 ,探讨该方改善学习记忆的作用机制。方法 :采用D 半乳糖致亚急性衰老合并Meynert核损毁的AD大鼠模型 ,实验分为青年组、老年组、模型组、石杉碱甲组、补肾益智方低剂量组、补肾益智方高剂量组 ,用电镜结合体视学分析方法观察各组大鼠前额皮皮及海马CA1区突触的形态结构。结果 :模型组突触的数密度(Nv)、面密度 (Sv)均显著低于青年组和老年组 (P <0 0 1) ;补肾益智方高、低两个剂量组均能显著增加AD模型大鼠突触的Nv和Sv(P <0 0 1) ,且高剂量组增加Nv的作用较低剂量组明显 (P <0 0 5 )。结论 :补肾益智方有促进新的突触形成 ,增加突触数量的作用 ,且对于AD模型大鼠前额皮质及海马CA1区突触数密度的增加有明显的量效关系。
Objective : To observe the effect of Bushen Yizhi Recipe on the synapse of Alzheimer’s disease (AD) model rats, and to explore the mechanism of this prescription to improve learning and memory. METHODS: AD rat models with subacute senescence and Meynert nuclear damage caused by D galactose were divided into young group, old group, model group, Huperzine A group, Bushen Yizhi low dose group and Bushen Yizhi Fang. In the high-dose group, the morphological structure of synapses in the prefrontal cortex and hippocampal CA1 region was observed by electron microscopy combined with stereological analysis. Results: The number density (Nv) and areal density (Sv) of the synapses in the model group were significantly lower than those in the young group and the old group (P < 0.01). Both the high and low doses of Bushen Yizhi were significantly increased. Synaptic Nv and Sv (P < 0.01) in the AD model rats, and the effect of increasing Nv in the high dose group was significantly lower (P <0 05). Conclusion: Bushen Yizhi Recipe promotes the formation of new synapses and increases the number of synapses, and has a significant dose-effect relationship for the increase of synaptic number density in the prefrontal cortex and hippocampal CA1 region in AD rats.
