目的探讨凋亡肿瘤细胞抗原负载的树突状细胞(DC)对喉癌细胞杀伤作用的影响。方法用细胞因子重组人白细胞介素4(rhIL-4)、重组人粒细胞巨噬细胞集落刺激因子(rhGM-CSF)与肿瘤坏死因子α(rhTNF-α)体外诱导扩增人外周血单个核来源的DC,6 d后将DC分为凋亡组、冻融组和未处理组;各组细胞继续培养4 h后,分别采用流式细胞术检测DC表面分子的表达、ELISA法检测干扰素γ(IFN-γ)的水平、MTT法评估各组DC刺激细胞毒性T淋巴细胞(CTL)增殖的能力及诱导CTL的杀伤活性。结果与其余各组相比,凋亡组DC表面分子与IFN-γ的表达明显增加(P<0.05),CTL的增殖能力与杀伤活性增强(P<0.05)。结论凋亡肿瘤细胞可以促进DC的分化与成熟,并能使DC产生高效而特异性的抗喉癌免疫应答。
Objective To investigate the effect of dendritic cells (DCs) loaded with apoptotic tumor cells on the cytotoxicity of laryngeal carcinoma cells. Methods Human rhIL-4, rhGM-CSF and rhTNF-α were used to induce the proliferation of human peripheral blood mononuclear cells DCs were divided into apoptotic group, frozen-thawed group and untreated group after 6 days. After cultured for 4 hours, DCs were detected by flow cytometry. The expression of interferon γ (IFN-γ) levels were measured by MTT method. The ability of DCs to stimulate proliferation of cytotoxic T lymphocytes (CTLs) and CTL-induced cytotoxicity were evaluated by MTT assay. Results Compared with the other groups, the expression of DCs and IFN-γ in apoptotic group were significantly increased (P <0.05), and the proliferation and cytotoxicity of CTL were enhanced (P <0.05). Conclusion Apoptosis of tumor cells can promote the differentiation and maturation of DC and make DC produce efficient and specific anti-laryngeal cancer immune response.
