目的:研究独活颗粒剂对Aβ1–40蛋白所诱发的AD模型大鼠免疫损伤的抑制作用,探讨独活延缓脑老化的作用机制。方法:采用Aβ1–40灌注法建立AD大鼠模型,分别用中药独活颗粒剂、西药吲哚美辛、0.9%的氯化钠溶液灌胃,选择ELISA双抗体夹心方法,检测各组大鼠血清中细胞因子IL-4、TNF-α的表达变化。结果:与假手术组比较,AD模型大鼠血清TNF-α的含量增加(P<0.01),IL-4的含量减少(P<0.01);与模型组比较,独活组大鼠血清TNF-α的含量减少(P<0.01),IL-4的含量增加(P<0.01)。结论:独活颗粒剂对Aβ1–40蛋白所诱发的老年性痴呆病(AD)模型大鼠的炎症反应具有抑制作用;独活颗粒剂通过调节炎症反应而具有明显的延缓脑老化作用。
OBJECTIVE: To study the inhibitory effect of Duowu Granule on immune injury induced by Aβ1-40 protein in AD model rats, and to explore the mechanism of action alone in delaying brain aging. Methods: A rat model of Alzheimer’s disease (AD) was established by perfusion with Aβ1-40. The rats were randomly divided into the following groups: Indomethacin, Indomethacin and 0.9% sodium chloride solution respectively. ELISA double antibody sandwich method was used to detect the serum In the changes of cytokines IL-4, TNF-α expression. Results: Compared with the sham operation group, the content of TNF-α in serum increased (P <0.01) and the content of IL-4 in AD model group decreased (P <0.01) (P <0.01), the content of IL-4 increased (P <0.01). Conclusion: Dove Capsule can inhibit the inflammatory reaction induced by Aβ1-40 protein in Alzheimer ’s disease (AD) model rats. Duowei granules can delay brain aging by regulating inflammatory reaction.
