目的 研究NK 92细胞对人卵巢癌细胞的体外杀伤活性以及放射线照射后对其杀伤活性的影响。方法 以体外培养人卵巢癌细胞系HO 8910为靶细胞 ,以NK 92的敏感细胞株K5 6 2作为对照 ,应用 4h51Cr释放法检测不同效靶比情况下体外培养NK 92细胞的杀伤活性 ,并应用医用电子直线加速器对NK 92细胞进行照射处理 (0、4 0 0cGy)后再检测其杀伤活性变化。结果 NK 92细胞未照射组 ,效靶比较低时 (1∶1、5∶1)对于HO 8910细胞杀伤率为 39%~4 5 % ,随效靶比升高 ,杀伤率随之上升 ,10∶1时杀伤率显著上升为 5 9% ,2 0∶1时杀伤率仅上升了 6 % ;对于K5 6 2细胞 ,杀伤率为 5 8%~ 71%。照射后 2d ,4 0 0cGy组NK 92细胞对HO 8910细胞不同效靶比的杀伤率与 0cGy组相比有所降低 ,其总体杀伤率为 4 2 %~ 6 2 % ,对K5 6 2细胞杀伤范围在 4 4 %~ 6 1%之间。结论 NK 92细胞对人卵巢癌细胞系HO 8910具有较高的杀伤活性 ,经放射线照射后仍保留一定的杀伤活性。
Objective To study the in vitro cytotoxic activity of NK-92 cells on human ovarian cancer cells and their cytotoxicity after irradiation. Methods Human ovarian cancer cell line HO 8910 was used as target cells and NK 92 sensitive cell line K5 6 2 as control. The cytotoxic activity of NK 92 cells cultured in vitro was detected by 4h51Cr release assay. The NK 92 cells were irradiated by medical electron linear accelerator (0, 400 cGy) before their killing activity was tested. Results The killing rate of NK-89 cells was 39% ~ 45% when the effective target ratio was low (1: 1, 5: 1) : 1, the killing rate increased significantly to 59%, and the killing rate increased only 6% at 20%. The killing rate of K562 cells ranged from 58% to 71%. At 2 days after irradiation, NK 92 cells in the 400 cGy group had a lower killing effect on different target ratios of HO 8910 cells compared with the 0 cGy group, with an overall killing rate of 42% -62%, killing K5 6 2 cells The range of 4 4% ~ 6 1%. Conclusion NK 92 cells have high cytolytic activity against human ovarian cancer cell line HO 8910 and still retain certain cytotoxicity after irradiation.
