目的探讨神经苷节脂特异性的唾液酸酶(Neu3)基因与人类结直肠癌的关系。方法用逆转录聚合酶链反应(RTPCR)方法检测8例结直肠癌组织和正常黏膜之间Neu3mRNA的表达差异;用硫巴比妥酸方法(TBA)检测32例结直肠癌组织和正常黏膜中唾液酸酶活性。结果在转录水平,8对结直肠癌组织和正常组织中,癌组织中Neu3mRNA的表达明显高于正常组织中Neu3mRNA的表达水平;在蛋白质翻译水平,32例结直肠癌组织中的唾液酸酶活性平均为10.60U/mg,32例正常黏膜组织中的唾液酸酶活性平均为5.51U/mg(P<0.05)。结论Neu3基因的编码产物神经苷节脂特异性的唾液酸酶与结直肠癌细胞的分化和生长密切相关。Neu3基因可能与人类结直肠癌有关。
Objective To investigate the relationship between the neuronal ganglioside-specific sialidase (Neu3) gene and human colorectal cancer. Methods The expression of Neu3 mRNA in 8 colorectal cancer tissues and normal mucosa was detected by reverse transcriptase polymerase chain reaction (RTPCR). The expressions of Neu3 mRNA in colorectal cancer tissues and normal mucosa were detected by thiobarbituric acid assay (TBA) Sialidase activity. Results At the transcriptional level, the expression of Neu3mRNA in cancer tissues was significantly higher than that in normal tissues in 8 pairs of colorectal cancer tissues and normal tissues. At the protein translation level, the sialidase activity in 32 cases of colorectal cancer tissues With an average of 10.60U / mg. The average sialidase activity in 32 normal mucosa tissues was 5.51U / mg (P <0.05). Conclusion The sirtuin, a gene encoding the Neu3 gene, is closely related to the differentiation and growth of colorectal cancer cells. Neu3 gene may be related to human colorectal cancer.
