目的探讨孕期膳食补锌对胎盘及母鼠小肠中IGF-II mRNA表达的影响。方法18只SD孕鼠随机分为正常对照组(ZC)和补锌组(ZS)。两组进食常规饲料,ZC组(n=9)饮用去离子水,ZS组(n=9)饮用补锌水(含锌1.26 mmol/L)。分别于孕9.5 d和孕17.5 d时取胎盘和小肠。以半定量聚合酶链反应(RT-PCR)测定胎盘和母鼠小肠中IGF-II mRNA的相对表达量。结果ZC组孕17.5 d时,胎盘IGF-II mRNA的相对表达量明显高于孕9.5 d(P<0.05),呈现出发育性增高。孕9.5 d和孕17.5 d时,ZS组胎盘及小肠IGF-II mRNA相对表达量均显著高于ZC组(P<0.05)。结论妊娠不同时期膳食锌摄入水平增高显著上向调控胎盘和母体小肠中IGF-II mRNA的表达。
Objective To investigate the effects of dietary zinc supplementation during pregnancy on the expression of IGF-II mRNA in the small intestine of placenta and maternal rats. Methods Twenty - eight SD pregnant rats were randomly divided into normal control group (ZC) and zinc supplementation group (ZS). The two groups consumed conventional diet. ZC group (n = 9) drank deionized water while ZS group (n = 9) consumed zinc supplemented water (1.26 mmol / L zinc). Placenta and small intestine were taken at 9.5 d of pregnancy and 17.5 d of pregnancy, respectively. Relative expression of IGF-II mRNA in placenta and maternal small intestine was determined by semi-quantitative polymerase chain reaction (RT-PCR). Results The relative expression of IGF-II mRNA in placenta of ZC group was significantly higher than that of pregnant group at 9.5 d (P <0.05) at 17.5 d of gestation, showing a developmental increase. At 9.5 and 17.5 d of pregnancy, the relative expression of IGF-II mRNA in the placenta and small intestine of ZS group was significantly higher than that of ZC group (P <0.05). Conclusion The increase of dietary zinc intake in different stages of pregnancy significantly up-regulated the expression of IGF-II mRNA in placenta and maternal small intestine.
