目的优化黄伞胞外多糖发酵培养基。方法在单因素实验的基础上,采用Plackett-Burman实验设计筛选影响多糖产量的培养基成分,然后用最陡爬坡路径逼近最大响应区域,最后通过Box-Behnken设计及响应面分析确定主要影响因素的最佳浓度。结果获得影响多糖产量的培养基成分为:麸皮、黄豆饼粉和磷酸二氢钾。通过Box-Behnken设计及响应面分析确定主要影响因素的最佳浓度为:麸皮11g/L、黄豆饼粉7.3g/L、磷酸二氢钾0.8g/L。结论利用响应面分析法优化黄伞胞外多糖发酵培养基是可靠的,具有实际的应用价值。
Objective To optimize the exopolysaccharide fermentation medium of Methods Based on the single factor experiment, Plackett-Burman experimental design was used to screen the components of the medium affecting the yield of polysaccharides. Then the steepest ascent path was used to approximate the maximum response area. Finally, the main influencing factors were determined by Box-Behnken design and response surface analysis The best concentration. Results The medium components that affected the yield of polysaccharides were bran, soybean meal and potassium dihydrogen phosphate. The optimum concentrations of the main influencing factors determined by Box-Behnken design and response surface analysis were: bran 11g / L, soybean meal 7.3g / L, potassium dihydrogen phosphate 0.8g / L. Conclusion Optimization of response surface methodology to extract exopolysaccharide fermentation medium is reliable and has practical value.
