目的:探讨加味木防己汤对佐剂性关节炎(AA)大鼠滑膜细胞基质金属蛋白酶(MMP)生成的影响及其可能的分子机制。方法:将Wistar大鼠随机分4组,每组10只,并应用弗氏完全佐剂诱发大鼠AA实验性模型,造模18d开始给药,第40天处死大鼠取膝关节滑膜组织体外进行原代细胞培养,通过明胶酶谱分析法检测加味木防己汤对AA大鼠滑膜细胞MMP-2和MMP-9生成的影响。结果:①正常组大鼠滑膜细胞仅产生MMP-2;②在TNF-α(10μg/L)存在下,正常组大鼠可表达MMP-9,AA组大鼠滑膜细胞MMP-2和MMP-9生成量增加;③加味木防己汤治疗组大鼠滑膜细胞,在无干预情况下MMP-2和MMP-9的表达量均显著降低,在TNF-α存在时MMP-9的表达量未见明显增加。结论:加味木防己汤不仅能抑制AA大鼠滑膜细胞生成MMP-2和MMP-9,同时也能抵抗TNF-α对AA大鼠滑膜细胞生成MMP-9的刺激作用。提示加味木防己汤可能具有调控类风湿关节炎发病的作用。
Objective: To investigate the effect of Jiawei Mufangfang on the production of matrix metalloproteinase (MMP) in synoviocytes of adjuvant arthritis (AA) rats and its possible molecular mechanism. METHODS: Wistar rats were randomly divided into 4 groups with 10 rats in each group. The rat model of AA was induced by Freund’s complete adjuvant and the model was established on day 18 of the injection. The synovial tissue of the knee joint was sacrificed on the 40th day. The primary cell culture was performed in vitro and the effect of Jiawei Mufang Decoction on the production of MMP-2 and MMP-9 in synoviocytes of AA rats was detected by gelatin zymography. RESULTS: 1 Synoviocytes in normal rats only produced MMP-2; 2 In the presence of TNF-α (10 μg/L), rats in normal group could express MMP-9, while those in AA group had MMP-2 and synoviocytes. The production of MMP-9 increased; 3 Syngastric cells in rats treated with Jia Fang Mu Fang Decoction were treated. In the absence of intervention, the expression levels of MMP-2 and MMP-9 were significantly decreased, and the expression of MMP-9 in the presence of TNF-α was increased. No significant increase in volume. Conclusion: Jiawei Mufang Decoction can not only inhibit the formation of MMP-2 and MMP-9 in synoviocytes of AA rats, but also resist the stimulatory effect of TNF-α on MMP-9 production by synoviocytes of AA rats. It is suggested that Jiafenmufang Decoction may have the effect of regulating the onset of rheumatoid arthritis.
