目的观察荞麦花叶发酵提取物(Extract of fermented buckwheat flower and leaf,EFBFL)对MIN6细胞促增殖,抗凋亡作用,为荞麦花叶进一步开发研究提供实验依据。方法以不同浓度EFBFL干预胰岛MIN6细胞,以高糖(含葡萄糖33.3 mmol/L)刺激作为诱导凋亡或抑制增殖因素。用cck-8试剂盒检测EFBFL对MIN6细胞增殖活性的影响;用流式细胞仪(Annexin V-FITC和PI双染法)检测EFBFL对胰岛细胞凋亡的影响。结果 EFBFL在16、32、64μg/ml浓度下对高糖和非高糖刺激的胰岛MIN6细胞增殖活性均有促进。与高糖对照组比较,32和64μg/ml的EFBFL对高糖诱导的胰岛细胞早期凋亡和晚期凋亡有明显抑制作用(P<0.01),有浓度效应关系。结论 EFBFL具有良好的促进MIN6细胞增殖和抑制高糖诱导的胰岛MIN6细胞的凋亡作用。
Objective To observe the effect of extract of fermented buckwheat flower and leaf (EFBFL) on the proliferation and anti-apoptosis of MIN6 cells, and provide experimental basis for the further development of buckwheat mosaic. Methods Different concentrations of EFBFL interfered with the islets of MIN6 cells with high glucose (containing 33.3 mmol / L glucose) as stimulation of induction of apoptosis or inhibition of proliferation. The effect of EFBFL on the proliferation of MIN6 cells was detected by cck-8 kit. The effect of EFBFL on the apoptosis of islet cells was detected by flow cytometry (Annexin V-FITC and PI double staining). Results EFBFL promoted the proliferation of MIN6 cells stimulated by high glucose and non-glucose at 16, 32 and 64μg / ml. Compared with the high glucose control group, EFBFL of 32 and 64μg / ml significantly inhibited the early apoptosis and late apoptosis induced by high glucose (P <0.01), and had a concentration-dependent effect. Conclusion EFBFL can promote the proliferation of MIN6 cells and inhibit the apoptosis of MIN6 cells induced by high glucose.
