目的:探讨心房颤动(房颤)患者心房肌乙酰胆碱兴奋钾电流(IK,Ach)通道基因和蛋白表达变化,以了解IK,Ach通道在房颤发生中的可能作用机制。方法:入选行心脏瓣膜病手术患者作研究对象,其中并发房颤者9例(房颤组),窦性心律者13例(对照组)。所有入选患者于开胸行心脏手术时切取右心耳组织,房颤组患者同期加取左心耳组织。用RT-PCR和Western-Blot检测不同心房肌组织中人Kir3.4基因及蛋白水平的表达。结果:房颤组患者IK,Ach通道蛋白mRNA表达水平及蛋白含量均较对照组患者减少(P<0.05)。结论:IK,Ach通道蛋白mRNA表达水平及蛋白含量较少可能是慢性房颤患者发病机制之一。
Objective: To investigate the changes of gene and protein expression of atrialkinese kinase (AK) channel in atrial fibrillation in patients with atrial fibrillation (Atrial fibrillation) in order to understand the possible mechanism of IK and Ach channels in atrial fibrillation. Methods: Patients undergoing valvular heart disease were enrolled in this study. Among them, 9 cases were complicated by atrial fibrillation (AF group), 13 cases were sinus rhythm (control group). All selected patients underwent cardiac surgery in the thoracotomy right atrial appendage, atrial fibrillation patients in the same period plus left atrial appendage. The expression of human Kir3.4 gene and protein in different atria were detected by RT-PCR and Western-Blot. Results: The mRNA and protein levels of IK and Ach channel in atrial fibrillation group were significantly lower than those in control group (P <0.05). Conclusion: The expression of IK and Ach channel protein mRNA may be one of the pathogenesis of chronic atrial fibrillation.
