LGT蛋白质组对肿瘤患者临床意义的首次观察

目的初步分析LGT(lostgoodwilltarget)蛋白质组基本临床意义指向。方法将2004年4月至2004年12月31日治疗的146例肿瘤患者用美国赛弗吉(Ciphergen)公司的蛋白质指纹仪及WCX2蛋白质芯片经SELDI检测技术(Surfaceenhancedlaserdesorption/ionization-timeofflight-massspectrometer)检测其血清。以在指纹图上,质荷比(M/Z)为11100+H至11900+H之间出现一峰簇样(cluster)的指纹标志为阳性诊断标准,结合检测次数,共分为6组,观察6组中的死亡率(观察工作于2005年3月20日结束)。结果LGT蛋白质组持续阳性组,死亡率100%;LGT蛋白质组持续阴性组,死亡率为0;LGT蛋白质组由阳性治疗后转成阴性组死亡率为0;LGT蛋白质疑似阳性组,死亡率9.5%;LGT蛋白质组仅查一次阳性组,死亡率17%;LGT蛋白质组查一次阴性组,死亡率1.5%。结论LGT蛋白质组持续阳性,会危及肿瘤患者的生命,由于它的出现距死亡发生有一定时间,所以它是一个早期发现肿瘤恶化、预测死亡发生的早期预警蛋白质组。它的持续阴性表达可以提示肿瘤患者病情稳定,近期没有恶化和死亡的发生,而其从阳性经治疗转阴也可达到此目的。因此,将其作为一种临床诊断平台,以预测肿瘤的发展,并提示相应治疗,对患者长期生存有明确的临床意义。 Objective To analyze the basic clinical significance of LGG (lostgoodwilltarget) proteome. Methods 146 cancer patients treated from April 2004 to December 31, 2004 were detected with protein fingerprint instrument and WCX2 protein chip of American Ciphergen company by SELDI detection technique (ionization-timeofflight-mass spectrometer) Its serum. CONCLUSIONS: The LGT proteome is consistently positive and will endanger the life of tumor patients. Since it appears for some time before death, it is an early warning proteome that early detection of tumor progression and death prediction. Its continued negative expression may prompt the stable condition of patients with cancer, no recent deterioration and death, and its positive from the treatment of negative can also achieve this goal. Therefore, as a clinical diagnostic platform to predict the development of the tumor, and prompt the corresponding treatment, patients with long-term survival has a clear clinical significance.
