研究乳香提取物提高小鼠免疫功能的机理与途径,探索乳香提取物对增强免疫功能的应用潜能。采用环磷酰胺制备免疫低下小鼠模型,以乳香提取物经灌胃给予免疫低下小鼠,连续10d。灌胃结束后取小鼠外周血采用荧光抗体染色并经流式细胞仪检测小鼠T细胞数量变化;取小鼠脾脏单个核细胞加抗CD3和CD28刺激并经3 H-TdR掺入试验,观察小鼠淋巴细胞增殖能力;取小鼠骨髓前体细胞与GM-CSF共培养,3 H-TdR掺入试验观察小鼠骨髓前体细胞增殖能力;ELISA方法检测小鼠血清中细胞因子。结果表明,给免疫低下模型小鼠口服乳香提取物后可有效增加小鼠总T细胞及CD4+T细胞数量:非服用组T细胞为12%±3%,服用组T细胞为28%±4%,两组相比具有显著差异(P<0.01);而CD4+T细胞从5%±2%(非服用组)增加至22%±3%(服用组),两组相比具有显著性统计学差异(P<0.01)。接受乳香提取物处理小鼠的T细胞对抗CD3和CD28刺激的增殖反应明显增高(P<0.05);IFN-γ、TNF-α、IL-12和IL-4水平上调;骨髓粒系细胞受GM-CSF刺激后增殖上调。因此,乳香提取物具有明显上调小鼠T细胞和Th1和Th2细胞数量和功能,其作用可能与刺激骨髓前体细胞增殖有关。
To study the mechanism and ways of Boswellia serrata extract to enhance immune function in mice and to explore the potential application of Boswellia extract to enhance immune function. Mice immunosuppressed with cyclophosphamide were prepared and immunosuppressed mice were administered with Boswellia serrata extract by gavage for consecutive 10 days. The peripheral blood of mice was stained with fluorescent antibody and the number of T lymphocyte was detected by flow cytometry after the end of gavage. The splenic mononuclear cells of mice were stimulated with anti-CD3 and CD28 and spiked with 3 H-TdR, The proliferation of mouse lymphocytes was observed. The mouse bone marrow precursor cells were co-cultured with GM-CSF. The proliferation of mouse bone marrow progenitor cells was observed by 3 H-TdR incorporation assay. The serum cytokines were detected by ELISA. The results showed that oral administration of Frankincense extract to mice with immunodeficiency model can effectively increase the total number of T cells and CD4 + T cells in mice: 12% ± 3% of untreated T cells and 28% ± 4% of T cells %, There was a significant difference between the two groups (P <0.01); while CD4 + T cells increased from 5% ± 2% (non-taking group) to 22% ± 3% (taking the group) Statistical difference (P <0.01). T cells treated with Boswellia serrata extract significantly increased the proliferative response to CD3 and CD28 stimuli (P <0.05); the levels of IFN-γ, TNF-α, IL-12 and IL-4 were up-regulated; -CSF stimulation increased proliferation. Therefore, frankincense extract significantly increased the number and function of T cells and Th1 and Th2 cells in mice, which may be related to stimulating the proliferation of bone marrow precursor cells.
