目的探讨齐墩果酸对氟伐他汀在大鼠体内的药动学影响。方法将16只健康大鼠随机分为单药组(5.0 mg·kg~(-1)氟伐他汀)和联合用药组(5.0 mg·kg~(-1)氟伐他汀+60 mg·kg~(-1)齐墩果酸),单药组和联合用药组分别灌胃空白溶剂和齐墩果酸5 d,每天1次,第6天2组均给予氟伐他汀灌胃。给药后不同时间点采血,LC-MS测定大鼠体内血药浓度,比较2组间主要的药动学参数。结果与单药组比较,联合用药组氟伐他汀主要药动学参数Cmax、AUC0-t显著上升,组间比较差异具有统计学意义(P<0.05)。结论联合应用齐墩果酸可能影响大鼠体内氟伐他汀的药动学特性。
Objective To investigate the effect of oleanolic acid on the pharmacokinetics of fluvastatin in rats. Methods Sixteen healthy rats were randomly divided into five groups: monotherapy group (5.0 mg · kg -1 fluvastatin) and combination therapy group (fluvastatin 5.0 mg · kg -1) +60 mg · kg -1 (-1) oleanolic acid). The single drug group and combination group were fed with blank solvent and oleanolic acid respectively for 5 days, once a day, and on the 6th day, both groups were given fluvastatin. Blood samples were collected at different time points after administration. LC-MS was used to determine the plasma concentration in rats. The main pharmacokinetic parameters were compared between the two groups. Results Compared with the single drug group, the main pharmacokinetic parameters Cmax and AUC0-t of fluvastatin in the combination group were significantly increased, the difference between the two groups was statistically significant (P <0.05). Conclusion The combination of oleanolic acid may affect the pharmacokinetics of fluvastatin in rats.
