目的:研究BTLA(B and Tlymphocyte attenuator)在T细胞上的表达和功能。方法:分离纯化人外周血T细胞,流式细胞术检测BTLA在不同T细胞亚群上的表达。进一步用CD3抗体分别和游离、包被以及抗Fc段二抗交联的BTLA激发单克隆抗体(mAb)或HVEM-Fc融合蛋白共同刺激T细胞活化,MTT检测细胞增殖。结果:BTLA组成性表达于CD3+、CD4+和CD8+T细胞表面。游离的BTLA mAb和HVEM-Fc融合蛋白均不能抑制CD3刺激的T细胞增殖,然而包被和交联后的BTLA mAb和HVEM-Fc融合蛋白则能够明显抑制T细胞增殖。结论:BTLA的活化能显著抑制CD3抗体所活化的T细胞的增殖。
Objective: To study the expression and function of BTLA (B and Tlymphocyte attenuator) on T cells. Methods: Purified human peripheral blood T cells, flow cytometry was used to detect the expression of BTLA on different T cell subsets. Further stimulation of T cell activation with CDLA antibody and BTLA-stimulated monoclonal antibody (mAb) or HVEM-Fc fusion protein, respectively, cross-linked with free, coated and anti-Fc secondary antibodies, respectively. Results: BTLA was constitutively expressed on the surface of CD3 +, CD4 + and CD8 + T cells. Both free BTLA mAb and HVEM-Fc fusion protein could not inhibit CD3-stimulated T cell proliferation, whereas coated and cross-linked BTLA mAb and HVEM-Fc fusion protein significantly inhibited T cell proliferation. Conclusion: Activation of BTLA significantly inhibits the proliferation of T cells activated by CD3 antibodies.
