目的建立一种测定灵芝孢子粉中多糖的单糖组成及含量的方法。方法采用沸水浴加热提取灵芝孢子粉中多糖,以2 mol·L-1三氟乙酸(TFA)水解多糖。采用离子色谱法进行测定,色谱柱为CarboPac PA10(4 mm×250 mm,10μm);淋洗液:2.5 mmol·L-1KOH溶液;柱温30℃;流速1.0 mL·min-1。结果 9种单糖在一定浓度范围内与色谱峰面积有良好的线性关系,加标回收率为85.6%~97.0%,RSD均小于3%;灵芝孢子粉中多糖主要由葡萄糖、半乳糖、甘露糖、木糖、岩藻糖等单糖组成。结论本实验所建立的方法分离效果好,灵敏度高,可用于灵芝孢子粉中多糖成分的分析研究。
Objective To establish a method for the determination of monosaccharide composition and content of polysaccharides in Ganoderma spore powder. Methods Polysaccharides in Ganoderma lucidum spore powder were extracted by boiling water bath and hydrolyzed polysaccharide with 2 mol·L-1 trifluoroacetic acid (TFA). The results of ion chromatography were as follows: CarboPac PA10 (4 mm × 250 mm, 10 μm); eluent: 2.5 mmol·L-1 KOH; column temperature was 30 ℃ and flow rate was 1.0 mL · min-1. Results Nine monosaccharides showed a good linear relationship with the chromatographic peak area in a certain concentration range. The recoveries were 85.6% ~ 97.0% with RSD less than 3%. The polysaccharides in Ganoderma lucidum spores were mainly composed of glucose, galactose, Sugar, xylose, fucose and other monosaccharides. Conclusion The method established in this experiment has good separation effect and high sensitivity, and can be used for the analysis of polysaccharides in Ganoderma lucidum spore powder.
