目的:探讨WNT信号通路受体FZD3mRNA及蛋白在大肠癌细胞及组织中的表达。方法:取正常大肠细胞株CCD18-Co、大肠癌细胞株SW480和SW620,以WNT信号通路PCR芯片筛选大肠癌细胞株中差异性表达的基因,并选择FZD3基因采用Taqman PCR法进行验证其在大肠癌细胞中的高表达,同时比较FZD3蛋白在大肠癌组织、腺瘤组织及癌旁正常组织的表达。结果:与大肠正常细胞株CCD18-Co比较,19个基因在大肠癌细胞株SW480和SW620中表达上调,其中FZD3基因为CCD18-Co的195倍和105倍;验证结果显示,与大肠正常细胞株CCD18-Co FZD3比较,FZD3 mRNA在SW480和SW620细胞株中较中升高820倍和622倍,验证了FZD3mRNA在大肠癌细胞株中的高表达;免疫组化结果显示,FZD3蛋白在大肠癌组织中表达为100%,大肠腺瘤中的表达为88%,而癌旁正常组织中仅为17.5%,与癌旁正常组织相比,FZD3蛋白在大肠癌组织中免疫组化分数高6.6倍(P<0.01);FZD3蛋白的高表达与肠癌TNM分期显著相关(P<0.005)。结论:FZD3可能在大肠癌的发生、发展中起到潜在癌基因的作用。
Objective: To investigate the expression of Wnt signaling pathway receptor FZD3 mRNA and protein in colorectal cancer cells and tissues. Methods: The normal colorectal cell line CCD18-Co and SW480 were used to screen the differentially expressed genes in colorectal cancer cell lines by WNT signaling pathway. The FZD3 gene was selected and verified by Taqman PCR in the large intestine The expression of FZD3 protein in colorectal cancer tissues, adenoma tissues and adjacent normal tissues was also compared. RESULTS: Compared with the normal colorectal cell line CCD18-Co, 19 genes were up-regulated in the colorectal cancer cell lines SW480 and SW620, and the FZD3 gene was 195-fold and 105-fold more than that of the CCD18-Co. The results showed that compared with the normal colorectal cell line FZD3 mRNA in SW480 and SW620 cell lines was 820- and 622-fold higher than CCD18-Co FZD3, which verified the high expression of FZD3 mRNA in colorectal cancer cell lines. Immunohistochemical results showed that FZD3 protein in colorectal cancer tissue , The expression of which was 88% in colorectal adenomas and only 17.5% in normal colorectal tissues. The FZD3 protein was 6.6 times higher in colorectal cancer tissues than in adjacent normal tissues P <0.01). The high expression of FZD3 protein was significantly associated with TNM staging of colorectal cancer (P <0.005). Conclusion: FZD3 may play a role in potential oncogenes in the occurrence and development of colorectal cancer.
