RNA与DNA的提取技术,从微量分离到大规模制备,已日趋完善。但有时样品数量较少如处理细胞标本时,则有必要同时提取RNA与DNA。目前一般采用CsCL超速离心获取具有不同密度离心速率的DNA和RNA。该法简单且收率高,但受设备条件限制。我们在提取人肺高转移巨细胞癌细胞系RNA用于构建cDNA文库时,将现有方法稍加改良,可以同时制备纯度较高的基因组DNA。
RNA and DNA extraction technology, from the micro-separation to large-scale preparation, has matured. However, in some cases, such as handling samples of cells, it is necessary to extract RNA and DNA simultaneously. Currently, CsCL ultracentrifugation is commonly used to obtain DNA and RNA with different densities of centrifugation. The method is simple and high yield, but subject to equipment conditions. When we extracted human lung metastatic giant cell carcinoma cell line RNA for the construction of a cDNA library, we slightly modified the existing method and prepared genomic DNA with high purity simultaneously.
