目的探讨创伤性脑损伤(TBI)大鼠脑组织周期依赖激酶(CDKs)Spy1的表达及细胞定位。方法 72只SD大鼠均分为九组:空白对照组、假手术组和7个TBI组(建立大鼠脑锐器切割伤模型后12h和1、3、5、7、14和28d共7个时间点组)。提取各组大鼠脑组织蛋白样品10μl,Westernblot和逆转录PCR的方法检测TBI后Spy1蛋白和Spy1mRNA表达。免疫荧光双标染色方法检测Spy1蛋白在脑组织中的细胞定位。结果 TBI后,Spy1蛋白及其mRNA的表达逐步升高,3d达高峰,此后逐渐降低恢复至接近正常水平。Spy1主要与星形胶质细胞和神经元之间存在共定位。结论大鼠TBI可以改变脑Spy1的表达,这种改变可能参与脑损伤后星形胶质细胞的活化和增殖过程。
Objective To investigate the expression and cellular localization of Spy1 in brain tissue of traumatic brain injury (TBI) rats. Methods Totally 72 SD rats were divided into nine groups: blank control group, sham operation group and 7 TBI group (12 rats were established at 12 h and 1, 3, 5, 7, 14 and 28 d Time point group). The protein expression of Spy1 and Spy1 in TBI rats were detected by Western blot and RT-PCR. Double immunofluorescence staining was used to detect the cellular localization of Spy1 protein in brain tissue. Results After TBI, the expression of Spy1 protein and its mRNA gradually increased and peaked at 3d, then gradually decreased and returned to the normal level. Spy1 is primarily co-localized with astrocytes and neurons. Conclusion TBI can change the expression of brain Spy1, which may be involved in the activation and proliferation of astrocytes after brain injury.
