目的观察酸性成纤维细胞生长因子(aFGF)对人膀胱癌细胞株EJ细胞的促增殖作用;以及酪氨酸蛋白激酶(TPK)抑制剂Genistein对aFGF诱导的EJ细胞增殖的抑制作用。方法以不同浓度的aFGF刺激EJ细胞,再对由aFGF引起的细胞增殖施加不同浓度的Genistein;以MTT法检测细胞的增殖程度,原位凋亡细胞检测(TUNEL)和流式细胞仪(FCM)检测Genistein对EJ细胞凋亡和细胞周期的影响。结果aFGF可以使EJ细胞增殖比明显增加,Genistein可引起EJ细胞增殖比明显下降,其程度均随浓度增高而增强;Genistein可以促进EJ细胞的凋亡,使细胞阻滞于G2M期。结论aFGF可以促进EJ细胞的增殖;Genistein可以诱导EJ细胞株的凋亡,对aFGF诱导的细胞增殖有抑制作用,可能为膀胱肿瘤的治疗提供新的尝试。
OBJECTIVE: To observe the effect of aFGF on proliferation of human bladder cancer cell line EJ and inhibition of aFGF-induced EJ cell proliferation by tyrosine kinase inhibitor Genistein. Methods EJ cells were stimulated with different concentrations of aFGF and different concentrations of Genistein were treated with aFGF. MTT assay was used to detect the proliferation, TUNEL and FCM, The effect of genistein on EJ cell apoptosis and cell cycle was examined. Results aFGF can significantly increase the EJ cell proliferation, Genistein can cause EJ cell proliferation decreased significantly, the extent increased with the concentration increased; Genistein EJ cells can promote apoptosis, the cells arrested in the G2M phase. Conclusions aFGF can promote the proliferation of EJ cells. Genistein can induce the apoptosis of EJ cells and inhibit the growth of aFGF-induced cells, which may provide a new try for the treatment of bladder cancer.
