目的 研究DPC4基因和神经细胞粘附分子(neuralcelladhesionmolecule,NCAM)分子表达的偶联性与胆管癌神经浸润转移的关系。方法 以RT PCR技术扩增 18例新鲜胆管癌标本RNA ,32 P标记DPC4 ,NCAM作探针 ,作斑点杂交 ,放射自显影图像分析确定 3种分子表达强度 ,建立DPC4和NCAM表达关系的数学模型。结果 DPC4基因表达强度随胆管癌恶性程度的增加而降低 ,NCAM则升高。数学关系模型为Y =0 .5 95 5 / (- 0 .0 3 3 9+X) ,显示两者呈负相关(P <0 .0 5 )。结论 DPC4基因可能与NCAM基因在功能上有负偶联性 ,DPC4可能通过某种信号转导途径调控NCAM基因的表达 ,从而影响胆管癌向神经浸润转移。
Objective To study the relationship between the coupling of DPC4 gene and neural cell adhesion molecule (NCAM) expression and neural invasion and metastasis of cholangiocarcinoma. METHODS: RT-PCR was used to amplify 18 specimens of fresh cholangiocarcinoma RNA, 32 P-labeled DPC4, NCAM as a probe, dot blot hybridization, and autoradiographic image analysis to determine the expression of three molecules and establish a mathematical model for the expression of DPC4 and NCAM. . Results The intensity of DPC4 gene expression decreased with the increase of malignancy of cholangiocarcinoma, while NCAM increased. The mathematical relationship model was Y = 0.595 5 / (- 0 .0 3 3 9 + X), showing a negative correlation between the two (P < 0.05). Conclusion The DPC4 gene may be negatively coupled to the NCAM gene. DPC4 may regulate the expression of NCAM gene through a signal transduction pathway, which may affect the invasion and metastasis of cholangiocarcinoma.
