目的探讨关键期内单眼形觉剥夺弱视大鼠模型反缝合治疗前后外侧膝状体突触后致密蛋白-95(PSD-95)的表达规律。方法14日龄大鼠随机为剥夺组、正常组及反缝合治疗组,经造模成功后取材,采用免疫组化和RT-PCR技术检测并分析各组大鼠外侧膝状体PSD-95蛋白和PSD-95mRNA表达水平的变化。结果同年龄段弱视组外侧膝状体PSD-95呈阳性神经元密度降低,PSD-95mRNA的表达减少(P<0.01);同年龄段反缝合治疗组外侧膝状体较弱视组PSD-95呈阳性神经元密度增加,PSD-95mRNA的表达增多(P<0.05),但仍低于正常组(P<0.05)。结论单眼剥夺及反缝合治疗影响大鼠外侧膝状体PSD-95的表达,提示PSD-95参与了视觉发育外侧膝状体可塑性的变化过程,是弱视发病的重要分子机制之一;验证了关键期内反缝合治疗对弱视外侧膝状体神经元功能状态的恢复作用,为临床弱视遮盖治疗提供了理论依据。“,”Objective To investigate the expression pattern of PSD-95 in the lateral geniculate nucleus of monocular form deprived rats before and after reverse suture treatment in the critical period. Methods The 14 day old rats were randomly divided into three groups:monocular deprivation group(MD),normal control group(NC)and reverse suture group (RS).After the model of rats had been made successfully,we measured the expression of PSD-95 protein and mRNA in the lateral geniculate nucleus of the different groups by using immunohistochemistry staining and reverse transcription-polymerase chain reaction (RT-PCR). Results The density of positive neurons and the ex-pression of PSD-95 mRNA in LGN of MD group was significantly much less than those in NC groups with the same age(P<0.01).The density of positive neurons and the expression of PSD-95 mRNA in RS group were higher than those of MD group(P<0.05),but still lower than those of NC group with the same age(P< 0.05). Conclusions Monocular deprivation and reverse suture treatment might influence the expression of PSD-95 in the lateral geniculate nucleus. The results implied that PSD-95 protein might participated in the plasticity of visual development. PSD-95 protein were one of the key molecules in the mechanism of amblyopia pathogenesis.The cover treatment may partially reverse expression of PSD-95 protein of deprived amblyopia neurons in the lateral geniculate nucleus during the critical period.
