目的:探讨维生素K3(VitK3)对腺样囊性癌细胞的凋亡诱导作用及其与As2O3的联合效果。方法:体外培养人腺样囊性癌细胞株(SACC-83),分别以VitK3、As2O3以及两者联合孵育细胞,采用四甲基偶氮唑蓝(MTT)法检测对腺样囊性癌细胞的抑制率,Annexin-V/Pl双染色流式细胞技术检测凋亡细胞的百分数,倒置显微镜及AO/EB染色观察细胞凋亡后的形态。采用SPSS 13.0软件包对数据进行方差分析和t检验。结果:体外实验表明,VitK3能抑制SACC-83细胞生长并呈剂量和时间依赖性,细胞死亡方式以凋亡为主。VitK3与As2O3联合应用时,具有协同作用。结论:VitK3能抑制腺样囊性癌细胞生长,并且与As2O3联合时有协同作用。
Objective: To investigate the apoptosis-inducing effect of vitamin K3 (VitK3) on adenoid cystic carcinoma cells and its combined effect with As2O3. Methods: The human adenoid cystic carcinoma cell line (SACC-83) was cultured in vitro. The cells were incubated with VitK3, As2O3 and the combination of both. The apoptosis of adenoid cystic carcinoma cells The percentage of apoptotic cells was detected by Annexin-V / Pl double staining flow cytometry. The morphology of apoptotic cells was observed by inverted microscope and AO / EB staining. Data were analyzed by ANOVA and t-test using SPSS 13.0 software package. Results: In vitro experiments showed that VitK3 could inhibit the growth of SACC-83 cells in a dose and time-dependent manner, and the cell death mode was mainly apoptosis. VitK3 and As2O3 combined with synergistic effect. Conclusion: VitK3 can inhibit the growth of adenoid cystic carcinoma cells and has a synergistic effect when combined with As2O3.
