研究发现,不少神经类疾病如老年痴呆症和体内淀粉样蛋白的聚集有关,这种蛋白纤维的聚集不仅和氨基酸的序列有关,而且和环境因素如金属离子、pH、温度等密切相关。本文利用原子力显微镜(AFM)观察了加入Fe3+离子后胰高血糖素的聚集过程,并用显微傅立叶变换红外光谱(Micro-FTIR)对聚集成的蛋白纤维的二级结构进行表征。实验结果表明:加入Fe3+离子后,胰高血糖素聚集成纤维的延迟时间大大推迟,同时形成纤维的表面形貌和二级结构都发生了很大变化,而且这种变化和加入的Fe3+离子浓度密切相关。
The study found that many neurological diseases such as Alzheimer’s disease and the accumulation of amyloid in vivo, the accumulation of this protein fiber not only with the amino acid sequence, but also with environmental factors such as metal ions, pH, temperature and other closely related. In this paper, the aggregation process of glucagon with Fe3 + ions was observed by atomic force microscopy (AFM), and the secondary structure of aggregated protein fiber was characterized by Micro-Fourier transform infrared spectroscopy (Micro-FTIR). The experimental results showed that the delay time of glucaggregation into fibrils delayed greatly after addition of Fe3 + ions, and the surface morphology and secondary structure of fibrils changed greatly. And this change and the addition of Fe3 + ion concentration closely related.
