目的运用生物信息学方法对日本血吸虫性别特异性候选基因SJCHGC06309(登录号为AY813032)进行分析,为该基因的进一步研究奠定基础。方法利用“数据库消减杂交”(Digital Differential Display,DDD)方法筛选出在日本血吸虫雄虫中特异表达的候选基因AY813032,使用生物信息学软件分析该基因的开放阅读框(ORF),细胞定位、蛋白序列等特征,并预测该基因的结构和功能。结果 AY813032蛋白由287个氨基酸编码,属于Cwf15/Cwc15结构域家族,定位于细胞核,有8个磷酸化位点,理论等电点(pI)为5.10,分子量为32.17 kDa,该蛋白在日本与曼氏血吸虫之间具有较高保守性。结论 AY813032基因为日本血吸虫性别特异性候选基因,可能与日本血吸虫性别发育相关。
Objective To analyze the sex-specific candidate gene SJCHGC06309 (accession number AY813032) of Schistosoma japonicum by bioinformatics method, and lay a foundation for further study of this gene. METHODS: AY813032, a candidate gene specifically expressed in male Schistosoma japonicum, was screened by DDD (Digital Differential Display) method. Bioinformatics software was used to analyze the open reading frame (ORF) of the gene. Cell localization , Protein sequences and other characteristics, and predict the structure and function of the gene. Results AY813032 protein encoded by 287 amino acids belongs to the Cwf15 / Cwc15 domain family. It locates in the nucleus and has 8 phosphorylation sites with a theoretical isoelectric point (pi) of 5.10 and a molecular weight of 32.17 kDa. Schistosoma schistosomiasis is highly conserved. Conclusion AY813032 gene is a sex-specific candidate gene of Schistosoma japonicum, which may be related to the gender development of Schistosoma japonicum.
