有效控制外植体褐变是茶树组织培养的关键技术。本文研究了外植体取材季节、切取方法、培养温度和不同抗氧化剂对外植体褐变率的影响。结果表明:春季材料的外植体污染率、褐变率比秋季的低,腋芽萌发率比秋季的高;外植体留部分叶柄、接种前用2.0 g?L-1 PVP溶液浸泡30 min、接种后在8℃避光培养16h可以有效降低外植体褐化率。春季材料、合适的外植体类型、培养温度及抗氧化剂的良好组合,有助于提高茶树腋芽初代培养的成功率。
Effective control of explants browning is the key technology of tissue culture of tea. In this paper, we studied the effects of explant selection season, cut method, culture temperature and different antioxidants on explant browning rate. The results showed that the pollutions and browning rates of explants in spring were lower than those in autumn and the germination rates of axillary buds were higher than those in autumn. Some petiole remained in explants and soaked in 2.0 g? L-1 PVP for 30 min before inoculation, After inoculation at 8 ℃ cultured in dark for 16h can effectively reduce the rate of explant browning. Spring material, suitable explant types, incubation temperature and a good combination of antioxidants help to improve the success rate of axillary bud primary culture of tea plant.
