目的测定天山岩黄芪总黄酮含量及其抗氧化活性。方法采用回流提取法,料液比为1∶30,70%乙醇70℃回流3h,提取天山岩黄芪中总黄酮,并以紫外分光光度法测定其中总黄酮含量;以Vc为阳性对照,测定天山岩黄芪总黄酮对DPPH·(超氧阴离子)与·OH(羟基自由基)的清除作用。结果回归方程为Y=11.663 X+0.007 9,r=0.999 6,线性范围:19.3~67.6μg/mL;方法精密度良好,RSD=1.2%;方法重现性良好,RSD=2.0%;在0~75min内样品稳定性良好,RSD=1.1%;样品回收率为99.9%,RSD=1.6%。天山岩黄芪中总黄酮含量为11.25mg/g,RSD=1.2%。天山岩黄芪总黄酮对DPPH·与·OH的清除率有浓度依赖性,当总黄酮浓度为50μg/mL时,对DPPH·的清除率为59.821%,对·OH的清除率为25.782%。结论方法简单易行、准确可靠,适用于天山岩黄芪总黄酮的含量测定,天山岩黄芪总黄酮具有较好的抗氧化活性。
Objective To determine the content of total flavonoids in Astragalus from Tianshan rock and its antioxidative activity. Methods The total flavonoids in Astragalus membranaceus collected from Tianshan Rock were extracted by reflux extraction, the ratio of solid to liquid was 1:30, the ethanol was refluxed at 70 ℃ for 3h, the content of total flavonoids was determined by UV spectrophotometry. Vc was used as a positive control to determine Tianshan Scavenging effect of total flavonoids from Astragalus membranaceus on DPPH · (superoxide anion) and · OH (hydroxyl radical). Results The regression equation was Y = 11.663 X + 0.007 9, r = 0.999 6, linear range: 19.3 ~ 67.6μg / mL. The method was accurate with a RSD of 1.2%. The reproducibility of the method was good with RSD 2.0% The sample stability was good within ~ 75min with RSD = 1.1%. The sample recovery was 99.9% with RSD = 1.6%. Tianshan Rock Astragalus total flavonoids content of 11.25mg / g, RSD = 1.2%. The total flavonoids of Astragalus from Tianshan Rock had a concentration-dependent effect on the scavenging rate of DPPH · and · OH. When total flavonoids concentration was 50μg / mL, the scavenging rate of DPPH · was 59.821% and · OH was 25.782%. Conclusion The method is simple, accurate and reliable. It is suitable for the determination of total flavonoids of Astragalus membranaceus in Tianshan rock. The total flavonoids of Astragalus membranaceus in Tianshan rock has good antioxidative activity.
