采用柱前衍生化高效液相色谱法测定欧李不同部位的γ-氨基丁酸(GABA)含量。色谱条件采用C18色谱柱(4.6mm×250 mm,5μm),以乙腈-0.1%乙酸溶液为流动相,检测波长为254 nm。γ-氨基丁酸在5~100μg·mL~(-1)范围内线性关系良好。结果表明,欧李不同部位的GABA含量差异较大,含量由高到低顺序为:当年生茎>芽>叶>花>花蕾>果肉>根,当年生茎中GABA含量最高(912.9μg·g~(-1)),根中GABA含量最低(263.3μg·g~(-1))。该方法操作简便、结果稳定、适合于欧李不同部位中γ-氨基丁酸含量测定,为欧李资源的综合开发提供了基础数据。
The contents of γ-aminobutyric acid (GABA) in different parts of the plum were determined by precolumn derivatization high performance liquid chromatography. The chromatographic conditions were as follows: C18 column (4.6 mm × 250 mm, 5 μm) with acetonitrile-0.1% acetic acid as the mobile phase at a detection wavelength of 254 nm. The linear relationship of γ-aminobutyric acid in the range of 5 ~ 100μg · mL ~ (-1) was good. The results showed that the content of GABA in different parts of the plum was significantly different from high to low in the order: stem> bud> leaf> flower> flower bud> pulp> root, and the highest content of GABA was 912.9 μg · g ~ (-1)), the lowest content of GABA in root (263.3μg · g -1). The method is simple, stable and suitable for the determination of γ-aminobutyric acid in different parts of European plum, providing the basic data for the comprehensive development of European plum resources.
