目的:探讨雷公藤内酯醇(triptolide,TP)对人绒癌细胞株JAR增殖、凋亡及凋亡调控蛋白BCL-2/BAX表达的影响。方法:以体外培养的人绒癌细胞株JAR为研究对象,MTT法检测TP对细胞增殖的抑制情况;TUNEL法观察TP作用于细胞后引起细胞凋亡情况,PCR和Western blot检测TP对凋亡调控蛋白BCL-2/BAX mRNA和蛋白表达的影响。结果:随着作用浓度的增加及时间延长,TP能明显抑制JAR细胞增殖并诱导其凋亡(P<0.05);并能下调凋亡抑制蛋白BCL-2的mRNA和蛋白的表达,上调凋亡促进蛋白BAX的mRNA和蛋白表达(P均<0.05)。结论:雷公藤内酯醇在体外能抑制人绒癌细胞株JAR增殖,诱导其凋亡;凋亡调控蛋白BCL-2/BAX表达水平的变化可能是TP诱导JAR细胞凋亡的重要机制。
AIM: To investigate the effects of triptolide (TP) on the proliferation and apoptosis of human choriocarcinoma cell line JAR and the expression of apoptosis regulatory protein BCL-2 / BAX. Conclusion: Triptolide can inhibit JAR proliferation and induce apoptosis of human choriocarcinoma cell line in vitro. The change of BCL-2 / BAX expression may be an important mechanism of TP-induced JAR cell apoptosis.
