以银杏(Ginkgo biloba L.)雄株叶片为试材,采用同源基因克隆及RACE-PCR方法,克隆到了类黄酮糖基转移酶(UDP-glycose:flavonoid glycosyltransferase,UFGT)基因GbUFGT的全长cDNA序列,其在GenBank中的注册号为JN640564.2。该基因编码区长1491bp,编码496个氨基酸。GbUFGT蛋白具有保守的PSPG基序、UDP-葡萄糖基转移酶和UDP-葡萄糖醛酸基转移酶结构域,与其他植物中的UFGT蛋白同源性较高。GbUFGT基因组序列与其cDNA序列相同,无内含子。半定量RT-PCR结果表明,GbUFGT在整个银杏叶片发育期均可较高水平的表达,且不同发育期表达水平无明显变化,属非发育时期特异性基因。
Using the male leaves of Ginkgo biloba L. as test material, the full-length cDNA of GbUFGT gene encoding UDP-glycose: flavonoid glycosyltransferase (UFGT) was cloned by homologous gene cloning and RACE-PCR Sequence, which is registered in GenBank as JN640564.2. The gene coding region 1491bp, encoding 496 amino acids. The GbUFGT protein has a conserved PSPG motif, UDP-glucosyltransferase and UDP-glucuronosyltransferase domains, with high homology to UFGT proteins in other plants. GbUFGT genomic sequence and its cDNA sequence the same, no introns. Semi-quantitative RT-PCR results showed that GbUFGT could express at a high level throughout the developmental stages of Ginkgo biloba leaves and had no significant changes at different developmental stages, which was a non-developmental stage specific gene.
