目的研究大夜关门中总黄酮的提取工艺及体内体外抗氧化性。方法通过超声波提取法提取大夜关门中黄酮类物质,测定羟基自由基清除能力及H_2O_2-FeSO_4自由基发生系统MDA生成的抑制率。结果大夜关门总黄酮最佳提取工艺为:提取温度为70℃,乙醇体积分数为70%,固液比为1∶20,提取时间为60 min。黄酮提取物可有效地清除羟自由基,且对小白鼠肝匀浆自氧化有明显的抑制作用。结论最佳工艺下黄酮提取率为19.43%,具有明显清除自由基及体内抗氧化功能,其医药保健价值有待进一步开发利用。
Objective To study the extraction process of total flavonoids and the anti-oxidation in vitro in large night close. Methods Flavonoids were extracted by ultrasonic extraction and the scavenging capacity of hydroxyl radical and inhibition of MDA production by H 2 O 2 -FeSO 4 free radical were determined. Results The optimal extraction process of total flavonoids from the big night was as follows: the extraction temperature was 70 ℃, the volume fraction of ethanol was 70%, the solid-liquid ratio was 1:20 and the extraction time was 60 min. Flavonoids extract can effectively scavenge hydroxyl radicals and inhibit the autoxidation of liver homogenates in mice. Conclusion The best extraction rate of flavonoids is 19.43%, which has obvious function of eliminating free radicals and anti-oxidation in vivo. Its medicinal health value needs to be further exploited.
