目的研究钴60灭活制备贾第鞭毛虫孢囊标准体的方法,并对孢囊保存条件进行初探。方法选取不同剂量(0~1200 Gy)钴60-γ射线对贾第鞭毛虫活体孢囊进行辐照,采用荧光染色方法判定辐照灭活效果,同时将灭活后的贾第鞭毛虫置于2.5%重铬酸钾中4℃保存,观察其在灭活后5个月内的变化。结果当钴60-γ射线辐照剂量为900 Gy时灭活率达到91.44%±1.64%,此时贾第鞭毛虫卵囊完整,抗原性保持完好,符合《生活饮用水卫生标准》要求。保存期为4个月时,细胞总数量、灭活率、细胞形态等均符合实验需求。结论 900 Gy钴60-γ射线辐照可作为贾第鞭毛虫孢囊灭活的有效方法,贾第鞭毛虫在2.5%重铬酸钾中4℃低温保存期限为4个月。
OBJECTIVE To study the method for the preparation of the standard of Giardia cysts by cobalt 60 inactivation and to study the conditions of cyst preservation. Methods Different dosages (0 ~ 1200 Gy) cobalt 60-γ ray were used to irradiate the cysts of Giardia lamblia. The inactivation effect was determined by fluorescence staining, and the inactivated Giardia was inoculated 2.5% potassium dichromate stored at 4 ℃, observe its changes within 5 months after inactivation. Results The inactivation rate reached 91.44% ± 1.64% when the dose of cobalt 60-γ-ray was 900 Gy. At this time, the oocysts of Giardia were intact and their antigenicity remained intact, which accorded with the requirements of Hygienic Standard of Domestic Drinking Water. Storage period of 4 months, the total number of cells, inactivation rate, cell morphology, etc. are in line with experimental needs. Conclusion 900 Gy Cobalt 60-γ-ray irradiation can be used as an effective method to inactivate cysts of Giardia lamblia. The duration of giardiasis storage at 4 ℃ in 2.5% potassium dichromate is 4 months.
