目的探讨急性冷应激对大鼠糖皮质激素(glucocorticoids,GCs)分泌及其受体(glucocorticoid receptor,GR)在外周血淋巴细胞中表达的影响。方法将雄性Wistar大鼠40只随机分为常温饲养组和急性冷应激6、12、24 h组,每组10只。试验大鼠经冷刺激后,收集大鼠血液,分离血清和外周血淋巴细胞,采用ELISA法检测各试验组大鼠血清中糖皮质激素皮质酮(corticosterone,CORT)浓度,q RT-PCR和Western blot法检测大鼠外周血淋巴细胞中GR基因m RNA和蛋白的表达水平。结果急性冷应激各组大鼠血清中CORT浓度均显著高于常温饲养组(P<0.01);急性冷应激各组大鼠外周血淋巴细胞中GR基因m RNA和蛋白表达水平均显著低于常温饲养组(P<0.05或<0.01)。结论急性冷应激可促使试验大鼠血清CORT浓度持续升高,该过程伴随着大鼠外周血淋巴细胞中GR表达水平的持续降低。
Objective To investigate the effects of acute cold stress on the expression of glucocorticoid receptor (GR) in peripheral blood lymphocytes of rats with glucocorticoids (GCs). Methods Forty male Wistar rats were randomly divided into normal temperature feeding group and acute cold stress 6,12,24 h group, 10 rats in each group. After cold stimulation, the blood of rats was collected and the serum and peripheral blood lymphocytes were separated. The concentrations of corticosterone (CORT), glucocorticoid (CORT), q RT-PCR and Western blot was used to detect the mRNA and protein expression of GR gene in peripheral blood lymphocytes of rats. Results The concentrations of CORT in serum of acute cold stress group were significantly higher than those of normal temperature group (P <0.01). The levels of m RNA and protein of GR gene in peripheral blood lymphocytes of acute cold stress group were significantly lower Feeding at room temperature (P <0.05 or <0.01). Conclusion Acute cold stress can promote the continuous increase of serum CORT concentration in rats, which is accompanied by the continuous decrease of GR expression in peripheral blood lymphocytes of rats.
