目的利用流式细胞仪技术分析比较基因敲入(KIMAP)和转基因(TGMAP)小鼠前列腺癌模型。方法对来源于KIMAP和TGMAP的肿瘤标本进行了流式细胞仪分析,并与临床前列腺癌进行了比较。结果来源于TGMAP(n=8例)和KIMAP(n=9例)的小鼠前列腺癌标本流式细胞仪分析显示,多数KIMAP肿瘤为双倍体,与临床相近,而所有TGMAP肿瘤均为非整倍体。结论由于KIMAP模型成功地模仿了人类前列腺癌特征,在前列腺癌临床前期研究中具有潜在的应用价值。
OBJECTIVE: To use flow cytometry (FCM) to compare the prostate cancer model of mice with gene knock-in (KIMAP) and transgenic (TGMAP) mice. Methods Tumor specimens from KIMAP and TGMAP were analyzed by flow cytometry and compared with clinical prostate cancer. Results Flow cytometric analysis of mouse prostate cancer specimens from TGMAP (n = 8) and KIMAP (n = 9) showed that most of the KIMAP tumors were diploid and similar to the clinical counterpart, whereas all TGMAP tumors were non- Euploid Conclusion Because KIMAP model successfully mimics the characteristics of human prostate cancer, it has potential value in the preclinical study of prostate cancer.
