研究枇杷叶中科罗索酸超声提取最佳工艺条件。在单因素试验的基础上,选定甲醇体积分数、料液比和时间作为影响因素,以科罗索酸提取率为评价指标。通过3因素3水平Box-Behnken中心组合试验,建立科罗索酸提取率的二次多项式回归方程,经响应面回归分析得到优化组合条件。结果表明,超声波法提取枇杷叶中科罗索酸的最佳工艺条件为:甲醇体积分数87.8%,料液比1:26(g/mL),超声时间34min。枇杷叶中科罗索酸的一次实际提取率可达0.525%,与理论预测值吻合较好,说明该优化方法可行。
To study the optimum conditions of ultrasonic extraction of corosolic acid in loquat leaves. On the basis of single factor test, methanol volume fraction, solid-liquid ratio and time were selected as influential factors, and the extraction rate of corosolic acid was taken as the evaluation index. The quadratic polynomial regression equation of corosolic acid extraction rate was established by the Box-Behnken combination test of 3-factor-3 level. The optimal combination conditions were obtained by response surface regression analysis. The results showed that the optimum conditions for the extraction of corosolic acid from loquat leaves by ultrasonic wave were as follows: methanol volume fraction 87.8%, solid-liquid ratio 1:26 (g / mL) and ultrasonic time 34 min. The actual extraction rate of corosolic acid in loquat leaves up to 0.525%, which is in good agreement with the theoretical predictions, indicating that the optimization method is feasible.
