目的观察酪酪肽(PYY)对体外培养的人胰腺癌Miapaca-2细胞的生长抑制作用及Y受体在胰腺癌细胞的表达。方法不同浓度的PYY分别作用于体外常规培养Miapaca-2细胞,采用MTT比色法检测细胞增殖情况,光镜观察细胞形态变化。RT-PCR法检测胰腺癌细胞Y受体亚型Y1、Y2、Y5 mRNA表达。结果 PYY对Miapaca-2细胞生长有明显的抑制作用,呈剂量依赖性,与对照组相比差异有统计学意义(P<0.01);胰腺癌Miapaca-2细胞表达Y1、Y2和Y5受体,Y2受体表达较强。结论 PYY对体外培养的胰腺癌细胞有明显的抑制作用,胰腺癌Miapaca-2细胞主要表达Y2受体。
Objective To observe the inhibitory effect of PYY on the growth of human pancreatic cancer Miapaca-2 cells and the expression of Y receptor in pancreatic cancer cells. Methods Different concentrations of PYY were used to culture Miapaca-2 cells in vitro. The proliferation of Miapaca-2 cells was detected by MTT colorimetric assay. The morphological changes of cells were observed by light microscope. The mRNA expression of Y receptor subtypes Y1, Y2 and Y5 in pancreatic cancer cells was detected by RT-PCR. Results PYY had a significant inhibitory effect on the growth of Miapaca-2 cells in a dose-dependent manner, which was significantly different from that of the control group (P <0.01). The expression of Y1, Y2 and Y5 receptors in Miapaca- Y2 receptor expression is strong. Conclusion PYY can significantly inhibit the proliferation of pancreatic cancer cells in vitro. The pancreatic cancer Miapaca-2 mainly expresses Y2 receptor.
