目的:了解局部注射重组人α肿瘤坏死因子(rhTNF-α)对牛骨形态发生蛋白(bBMP)在小鼠体内异位诱导成骨的影响。方法:采用X线检查成骨面积图像分析、碱性磷酸酶活性测定、钙含量测定和组织学检查等方法,对局部注射不同剂量rhTNF-α后bBMP诱导成骨活性的变化进行评价。结果:①bBMP5mg植入小鼠肌袋内可诱发一个完整的骨发育过程,导致异位骨软骨组织发生;②在bBMP植入区注射rhTNF-α后,bBMP诱导成骨活性受到干扰,表现为成骨活性随rhTNF-α注射剂量的增加而减小;③rhTNF-α使bBMP诱导的间充质细胞分化成骨过程受到影响,导致成骨时间延长或完全阻滞。结论:rhTNF-α在小鼠体内可削弱bBMP的异位诱导成骨能力并抑制间充质细胞分化成骨
OBJECTIVE: To investigate the effect of rhTNF-α injected into bovine bone morphogenetic protein (bBMP) induced ectopic osteogenesis in mice. Methods: The changes of bBMP-induced osteogenic activity after local injection of different doses of rhTNF-α were evaluated by X-ray examination of the image area of osteoblasts, alkaline phosphatase activity, calcium content and histological examination. Results: ①BBMP5mg implanted into the muscle pouch of mice could induce a complete process of bone development, resulting in ectopic osteochondral tissue. ②In the bBMP-implanted area, the osteogenic activity induced by bBMP was disturbed Bone activity decreased with the increase of the dosage of rhTNF-α; ③ rhTNF-α affected the differentiation of bBMP-induced mesenchymal cells into osteoblasts, resulting in prolonged or complete osteogenic time. CONCLUSION: rhTNF-α attenuates ectopic osteogenesis of bBMP and inhibits mesenchymal cell differentiation into osteoblasts in mice
