目的:观察与探讨芪冬活血饮对急性肺损伤(ALI)大鼠肺表面活性蛋白A(SP-A)在肺与大肠等组织表达影响,并且对SP-A在肺与大肠表达的相关性进行分析,从而为中医“肺与大肠相表里”理论提供实验依据。方法:SD大鼠60只,随机分为6组,采用气道内滴入脂多糖(LPS)制备ALI模型,各组均分别应用相应药物进行干预。所有大鼠均在造模后24h处死采集标本,采用ELISA和免疫组化法检测各组织SP-A表达,并记录肺湿干重比及进行HE染色光镜下观察肺损伤程度。结果:造模后24h,肺、大肠、支气管肺泡灌洗液(BALF)及血清中SP-A表达均下降。芪冬活血饮可以显著促进肺组织及血清中SP-A含量(P<0.05),减轻肺水肿及肺组织病理学损害。芪冬活血饮同大黄合用,可以促进ALI大鼠肺和大肠组织SP-A表达,提高BALF及血清中SP-A含量,减轻肺水肿及肺组织病理学损害。通过相关性分析,发现肺组织SP-A表达同BALF、大肠及血清表达呈正相关,但与大肠相关最密切(r=0.615)。结论:芪冬活血饮可以促进肺组织、大肠、BALF及血清中SP-A表达,减肺水肿,改善肺组织病理学损害,并且同大黄具有协同作用;经相关性分析,肺组织SP-A表达与大肠相关最密切。
Objective: To observe and explore the effect of Qidonghuoxue Yin on the expression of pulmonary surfactant protein A (SP-A) in the lung and large intestine of rats with acute lung injury (ALI) and the correlation between SP-A expression in lung and large intestine For analysis, so as to provide experimental evidence for TCM “theory of lung and large intestine phase”. Methods: Sixty Sprague Dawley rats were randomly divided into 6 groups. ALI model was induced by lipopolysaccharide (LPS) intratracheal instillation, and each group was given corresponding drugs for intervention. All rats were sacrificed 24 hours after model establishment. The expression of SP-A in each tissue was detected by ELISA and immunohistochemistry. The wet / dry weight ratio of lung was recorded and the degree of lung injury was observed by HE staining. Results: The expression of SP-A in lung, large intestine, bronchoalveolar lavage fluid (BALF) and serum was decreased 24 h after modeling. Qi Dong Huoxue Yin can significantly promote the lung tissue and serum SP-A content (P <0.05), reduce pulmonary edema and lung tissue pathology. Qi Dong Huoxue Yin combined with rhubarb, can promote ALI rat lung and colorectal tissue SP-A expression, increase the BALF and serum SP-A content, reduce pulmonary edema and lung tissue pathology. Correlation analysis showed that the expression of SP-A in lung tissue was positively correlated with the expression of BALF, large intestine and serum, but was most closely related to the large intestine (r = 0.615). Conclusion: Qidudonghuoxue Decoction can promote the expression of SP-A in lung tissue, large intestine, BALF and serum, reduce pulmonary edema, improve lung pathological damage and synergistic effect with rhubarb. Through correlation analysis, SP-A The expression is most closely related to the large intestine.
