目的探究微小RNA-21(miRNA-21)和蛋白磷酸酶2A癌性抑制因子(CIP2A)在卵巢癌组织中的表达及其与病理因素的关系。方法选取2011年3月至2015年3月间收治的81例卵巢癌患者,同时收集40例良性卵巢肿瘤患者作为对照,检测并比较各组患者的miRNA-21和CIP2A表达水平。结果卵巢癌组织中miRNA-21相对表达量为5.062±1.205,明显高于良性卵巢肿瘤组的1.145±0.232,差异有统计学意义(P<0.05)。卵巢癌患者血清中miRNA-21相对表达量为25.123±29.822,明显高于良性卵巢肿瘤组(0.901±1.232)和正常人(1.102±0.735),差异均有统计学意义(P=0.002)。Ⅱ~Ⅳ期卵巢癌组织中CIP2A表达率(Ⅱ期为38.1%、Ⅲ期为42.9%、Ⅳ期为62.5%)明显高于Ⅰ期(20.0%),差异有统计学意义(P<0.05)。低分化肿瘤中CIP2A高表达率(60.9%)明显高于组织学中、高分化肿瘤(18.4%、15.0%),差异有统计学意义。在正常卵巢组织中,CIP2A为低表达(30.0%)或不表达(70.0%)。结论血清及组织中miRNA-21的高表达与卵巢癌患者的关系确切,肿瘤组织中CIP2A的高表达与卵巢癌分期及组织学分级关系密切。miRNA-21和CIP2A检测可以作为早期诊断卵巢癌的有效指标。
Objective To investigate the expression of microRNA-21 (miRNA-21) and protein phosphatase 2A (CIP2A) in ovarian cancer and its relationship with pathological factors. Methods Eighty-one patients with ovarian cancer who were treated between March 2011 and March 2015 were enrolled in the study. Forty patients with benign ovarian tumors were collected as control. The expression of miRNA-21 and CIP2A in each group were detected and compared. Results The relative expression of miRNA-21 in ovarian cancer was 5.062 ± 1.205, which was significantly higher than that in benign ovarian tumor (1.145 ± 0.232, P <0.05). The relative expression level of miRNA-21 in ovarian cancer patients was 25.123 ± 29.822, which was significantly higher than that in benign ovarian tumors (0.901 ± 1.232) and normal people (1.102 ± 0.735) (P = 0.002). The expression rates of CIP2A in stage Ⅱ ~ Ⅳ ovarian cancer were 38.1% in stage Ⅱ, 42.9% in stage Ⅲ and 62.5% in stage Ⅳ, which were significantly higher than those in stage Ⅰ (20.0%) (P <0.05) . The poorly differentiated tumors CIP2A high expression rate (60.9%) was significantly higher than in histological, well-differentiated tumors (18.4%, 15.0%), the difference was statistically significant. In normal ovarian tissue, CIP2A is either low (30.0%) or not (70.0%). Conclusion The overexpression of miRNA-21 in serum and tissues is correlated with ovarian cancer patients. The high expression of CIP2A in tumor tissues is closely related to the staging and histological grade of ovarian cancer. miRNA-21 and CIP2A detection can be used as an effective indicator of early diagnosis of ovarian cancer.
