目的 探讨微波抗原修复对CD44s和CD44v6抗原的影响及其在颅内转移瘤及胶质母细胞瘤中的表达与这些肿瘤侵袭、转移之间的关系。方法 应用微波 -0 1%TritonX -10 0 (MW -T)和微波 -柠檬酸缓冲液 (MW -CA)修复抗原 ,微波 -LSAB免疫组化染色检测CD44s和CD44v6在 2 0例正常脑组织、3 5例脑胶质母细胞瘤和 3 0例颅内转移瘤中CD44的表达情况。结果 MW -CA和MW -T抗原修复处理的IOD显著高于未修复组的IOD值 (P <0 0 1) ;2 0例正常脑组织中CD44s和CD44v6表达均为阴性 ;3 5例脑胶质母细胞瘤CD44s阳性表达率为 10 0 % (3 5 /3 5 ) ,CD44v6表达为阴性 ;3 0例颅内转移瘤中CD44s阳性表达率为 86 7% (2 6/3 0 ) ,CD44v6阳性表达率为 66 7% (2 0 /3 0 )。CD44v6在颅内转移瘤与脑胶质母细胞瘤的表达差异有显著性 (P <0 0 1)。结论 微波抗原修复处理可以显著提高CD44s和CD44v6的IOD值 ;脑胶质母细胞瘤中CD44V6的表达可能与其脑内侵袭过程有关 ,无CD44v6表达可能与其很少发生颅外转移有关 ,并有可能成为颅内转移瘤诊治的有用指标之一。
Objective To investigate the effect of microwave antigen repair on CD44s and CD44v6 antigens and its relationship with the invasion and metastasis of intracranial metastases and glioblastomas. Methods The antigen was repaired by microwave-TritonX-10 (MW-T) and microwave-citric acid buffer (MW-CA). The expressions of CD44s and CD44v6 in 20 normal brain tissues were detected by microwave- 35 cases of brain glioblastoma and 30 cases of intracranial metastases CD44 expression. Results The IOD values of MW-CA and MW-T antigens were significantly higher than those of the untreated group (P <0.01). The expressions of CD44s and CD44v6 in 20 normal brain tissues were negative. The positive rate of CD44s in the blastocyst was 10 0% (3 5/3 5) and the expression of CD44v6 was negative. The positive rate of CD44s in 30 cases of intracranial metastases was 86 7% (26/3 0) The positive rate was 66.7% (20/30). There was a significant difference in the expression of CD44v6 between intracranial metastases and glioblastoma (P <0.01). Conclusion Microwave antigen repair can significantly increase the IOD value of CD44s and CD44v6; CD44V6 expression in brain glioblastoma may be related to the process of brain invasion, no CD44v6 expression may be associated with its rare occurrence of extracranial metastasis, and may become One of the useful indicators of diagnosis and treatment of intracranial metastases.
