目的 研究IL 5在人类鼻息肉组织中的含量、分布和表达 ,探讨其在嗜酸粒细胞聚集微环境控制中的意义和鼻息肉病的概念。方法 采用ELISA法和免疫组化ABC法检测IL 5在 4 0例鼻息肉患者鼻息肉组织中的含量和表达 ,并以鼻息肉患者和健康鼻下鼻甲粘膜作对照。结果 ①鼻息肉组织中IL 5的含量高于鼻息肉患者和健康鼻下鼻甲粘膜者 (P <0 0 5 ) ,后两者之间差异无显著性 (P >0 0 5 ) ;伴有变应性鼻炎鼻息肉组织中IL 5的含量高于不伴有变应性鼻炎者 (P <0 0 5 ) ;鼻息肉组织中IL5的含量与患者的年龄、性别无关 ;②IL 5在 80 1%的嗜酸粒细胞中表达 ,仅有 3 7%的淋巴细胞和 /或中性粒细胞表达IL 5 ,其在上皮细胞中则不表达 ;在所有表达IL 5的细胞中 90 9%为嗜酸粒细胞 ;IL 5在鼻息肉组织的嗜酸粒细胞中的表达强于鼻息肉患者和健康鼻下鼻甲粘膜者 (P <0 0 5 ) ,其在后两者中的表达差异无显著性 (P >0 0 5 ) ;IL 5在伴有变应性鼻炎鼻息肉组织的嗜酸粒细胞中的表达强于不伴有变应性鼻炎者 (P <0 0 5 ) ;IL 5在鼻息肉组织和下鼻甲粘膜的淋巴细胞和中性粒细胞中表达差异均无显著性 (P >0 0 5 ,P >0 0 5 )。结论 IL 5是鼻息肉组织中嗜酸粒细胞病理学机理中的关键细胞因子
Objective To study the content, distribution and expression of IL - 5 in human nasal polyps and to explore the significance of IL - 5 in the control of eosinophilic microenvironment and the concept of nasal polyposis. Methods The content and expression of IL-5 in nasal polyps of 40 patients with nasal polyps were detected by ELISA and immunohistochemical ABC method, and the nasal polyps were compared with healthy nasal turbinate mucosa. Results ① The content of IL-5 in nasal polyps was higher than that in nasal polyps and healthy nasal mucosa (P <0 05). There was no significant difference between the two groups (P 0 05) The content of IL 5 in nasal polyps should be higher than those without allergic rhinitis (P <0 05). The content of IL 5 in nasal polyps was not related to the age and sex of the patients. Of eosinophils, only 37% of lymphocytes and / or neutrophils express IL5, which is not expressed in epithelial cells; 909% of all cells expressing IL5 are eosinophils The expression of IL-5 in eosinophils of nasal polyps was stronger than that in nasal polyps and healthy nasal mucosa (P <0.05), and there was no significant difference in the expression of IL-5 between the latter two P> 0.05). IL-5 expression was higher in eosinophils with allergic rhinitis and nasal polyps than those without allergic rhinitis (P <0.05) There were no significant differences in the expression of lymphocytes and neutrophils between the tissue and inferior turbinate mucosa (P> 0.05, P> 0.05). Conclusion IL 5 is a key cytokine in eosinophilic pathology in nasal polyps
